To assess the effect of various culture media on the preservation of human gut microbiota, we examined Gut Microbiota Medium (GMM), Schaedler Broth (SM), Fermentation Medium (FM), and Carbohydrate Free Basal Medium (CFBM) in batch in vitro cultures treated with PMA. This study combined 16S rDNA sequencing (PMA-seq), untargeted LC-HR-MS/MS metabolomics, and supplementary GC-MS for short-chain fatty acid (SCFA) profiling. A feasibility study was undertaken to evaluate the viability of using pooled fecal samples (MIX) from fifteen healthy donors as inoculum before the experimental procedures, aiming to reduce the number of variables and enhance reproducibility in the in vitro cultivation tests. The results highlighted the suitability of pooling faecal samples for investigation into in vitro cultivation.
